Overview

NAT02 is an orally bioavailable, potent, and selective small molecule agonist of the growth hormone secretagogue receptor (GHSR-1a), also known as the ghrelin receptor. Originally discovered by AstraZeneca (as AZD8181) and subsequently licensed to Neurogastrx (as NG101) and Dr. Falk Pharma, NAT02 is being developed for the treatment of gastroparesis, including both diabetic and idiopathic forms. The drug acts as a prokinetic agent by mimicking the action of the endogenous hormone ghrelin, which stimulates gastric motility and accelerates gastric emptying. By activating GHSR-1a receptors in the gastrointestinal tract and enteric nervous system, NAT02 aims to alleviate the core symptoms of gastroparesis, such as nausea, vomiting, and early satiety. It is currently in Phase 2 clinical development as an oral alternative to injectable prokinetic therapies.
