Overview

Natalizumab is a recombinant humanized IgG4κ monoclonal antibody that targets the α4-integrin subunit on leukocytes. By binding to α4-integrin, it blocks the interaction of leukocytes with vascular cell adhesion molecules, thereby inhibiting their migration across the blood-brain barrier and into inflamed tissue. This mechanism reduces inflammation in diseases such as multiple sclerosis (MS) and Crohn's disease. Natalizumab is used as monotherapy for relapsing forms of MS—including clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease—and for inducing and maintaining remission in adults with moderately to severely active Crohn’s disease who have not responded adequately to conventional therapies or TNF-α inhibitors. It is administered by intravenous infusion every four weeks[1][2][3][8].
