This is a combination of two separate monoclonal antibody therapies: **Natalizumab** is a humanized IgG4κ monoclonal antibody that selectively binds to the α4-integrin component of adhesion molecules found on lymphocytes, monocytes, and eosinophils. It blocks the interaction of α4β1 integrin with VCAM-1 and α4β7 integrin with MAdCAM-1, preventing leukocyte migration across the blood-brain barrier into the central nervous system, thus reducing inflammation in multiple sclerosis. It is also used in Crohn's disease by blocking leukocyte adhesion to intestinal endothelium. **Rituximab** is a chimeric murine/human monoclonal antibody directed against the CD20 antigen found on B lymphocytes. It promotes B-cell lysis through complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity mechanisms.
