Overview

Natural house dust mite allergen extract is a biologic product used in allergen immunotherapy (AIT) for the treatment of house dust mite-induced allergic diseases, including allergic asthma and allergic rhinitis. The extract is typically derived from the house dust mite species *Dermatophagoides pteronyssinus* and *Dermatophagoides farinae*, containing a complex mixture of major and minor allergens such as Der p 1, Der p 2, Der f 1, and Der f 2. It is often formulated as a semi-depot preparation (adsorbed to aluminum hydroxide) for subcutaneous injection (SCIT). The mechanism of action involves inducing immunological tolerance by modulating the immune system's response to these allergens, shifting from a Th2-biased allergic profile to a Th1-biased or regulatory T cell (Treg) profile, and increasing the production of allergen-specific IgG4 blocking antibodies. Clinical trials, such as ChiCTR2500100575, evaluate accelerated up-dosing regimens to reach maintenance doses more efficiently in pediatric populations.
