NAV-240

NAV-240 is a clinical-stage tetravalent bispecific antibody designed to simultaneously target and inhibit OX40 ligand (OX40L) and tumor necrosis factor alpha (TNFα), two clinically validated drivers of inflammation in autoimmune diseases. By dual targeting these pathways, NAV-240 aims to downregulate both type 2 and non-type 2 inflammation, modulating adaptive immune responses as well as inflammatory cytokine signaling. This approach may offer improved efficacy over monotherapies for complex, heterogeneous autoimmune conditions with high unmet medical need. The drug is being developed by Navigator Medicines following in-licensing from IMBiologics; it was originally known as IMB101. Early clinical studies have demonstrated safety and tolerability with once-monthly dosing supported by pharmacokinetic data. Indications under investigation include hidradenitis suppurativa, rheumatoid arthritis, inflammatory bowel disease (including Crohn’s disease and ulcerative colitis), systemic sclerosis, and other difficult-to-treat autoimmune or inflammatory diseases[1][2][4][5][6][7][8][9].
