Overview
The Nav1.8 inhibitor developed by Niroda Therapeutics is a selective small-molecule antagonist of the voltage-gated sodium channel subtype 1.8 (NaV1.8). NaV1.8 is primarily expressed in peripheral sensory neurons (nociceptors) and plays a critical role in the initiation and propagation of action potentials in response to painful stimuli. By selectively blocking this channel, the drug aims to provide potent analgesia for acute and chronic pain conditions while avoiding the central nervous system side effects and addiction risks typical of opioids. Niroda is currently evaluating this asset in a Phase 3 combination trial (Trial ID: 2026-526867-39-00) alongside a NaV1.7 inhibitor (aneratrigine) to leverage the complementary roles of these two channels in pain signaling.
