Overview
Navarixin is an orally available small molecule antagonist of the C-X-C motif chemokine receptor 1 (CXCR1; interleukin-8 receptor alpha) and C-X-C motif chemokine receptor 2 (CXCR2; interleukin-8 receptor beta). It selectively binds to the extracellular domain of CXCR2, preventing activation by natural ligands like IL-8 and Gro-α. This inhibits neutrophil migration and pro-inflammatory cytokine release, key processes in inflammatory diseases. Navarixin has been investigated for asthma, chronic obstructive pulmonary disease (COPD), psoriasis, and cancers including metastatic castration-resistant prostate cancer, colorectal cancer, non-small cell lung cancer, and melanoma. Developed by Merck, preclinical studies show it can reduce neutrophil recruitment and inflammation in animal models of pulmonary disease.
