navepegritide

Navepegritide is an investigational, once-weekly subcutaneous **C-type natriuretic peptide prodrug** developed by Ascendis Pharma using its TransCon technology platform for **achondroplasia**. It is designed to provide sustained systemic exposure to active, unmodified CNP after subcutaneous administration, with low peak concentrations, thereby activating natriuretic peptide receptor-B and counteracting the overactive **FGFR3-driven** signaling that underlies impaired endochondral bone growth in achondroplasia. Clinical data described in phase 2 and phase 2b studies indicate improvements in annualized growth velocity, height z-scores, and measures of skeletal proportionality and lower-limb alignment, with a tolerability profile broadly comparable to placebo and low rates of injection-site reactions and hypotension. The program has also been studied in additional age groups and in long-term extension and combination studies with lonapegsomatropin.
