Overview

Navicixizumab is a bispecific monoclonal antibody that targets both the Notch ligand delta-like 4 (DLL4) and vascular endothelial growth factor A (VEGF-A). By simultaneously inhibiting DLL4-mediated Notch signaling and VEGF-driven angiogenesis, navicixizumab disrupts tumor blood vessel formation and maintenance, leading to potential anti-angiogenic and antineoplastic effects. The drug was originally developed by OncoMed Pharmaceuticals and is being further developed by Mereo BioPharma and Feng Biosciences. Navicixizumab has been investigated in clinical trials for several solid tumors, including ovarian cancer, colorectal cancer, breast cancer, fallopian tube cancer, gastric cancer, peritoneal cancer, with its highest development phase being Phase II for these indications[1][3][5]. Its dual mechanism aims to block tumor angiogenesis as well as inhibit tumor cell growth by targeting two key pathways involved in tumor progression[1][5].
