Overview

Navicixizumab + irinotecan is an investigational combination therapy under clinical evaluation for advanced solid tumors, particularly colorectal cancer. Navicixizumab is a bispecific humanized monoclonal antibody that targets both delta-like ligand 4 (DLL4), a key ligand in the NOTCH signaling pathway, and vascular endothelial growth factor (VEGF). By simultaneously inhibiting DLL4 and VEGF, navicixizumab disrupts tumor angiogenesis and overcomes resistance mechanisms associated with single-pathway inhibition. Irinotecan is a small molecule chemotherapeutic agent classified as a topoisomerase I inhibitor; it interferes with DNA replication in cancer cells, leading to cell death. The combination aims to enhance antitumor efficacy by targeting complementary pathways involved in tumor growth and survival. This regimen is being developed primarily for patients with metastatic or refractory colorectal cancer who have failed prior standard therapies[2][4][5][8].
