Drug intelligence / Profile preview
NAVIG (also known as MelVax) is an experimental therapeutic cancer vaccine developed by Altevax for the treatment of glioblastoma. The vaccine platform utilizes synthetic melanin nanoaggregates as an adjuvant to enhance immune responses. The formulation evaluated in the NAVIG-1 clinical trial (NCT06622434) consists of two peptide antigens targeting glioma-associated proteins: A52-Mel, which targets telomerase reverse transcriptase (TERT), and A49-Mel, which targets protein tyrosine phosphatase receptor type Z1 (PTPRZ1). These peptide formulations are adsorbed onto synthetic melanin and co-administered with Litenimod (Li28), a Toll-like receptor 9 (TLR9) agonist that acts as an immune adjuvant. By delivering these tumor-associated antigens in combination with melanin and a TLR9 agonist, NAVIG aims to stimulate a robust, antigen-specific CD4+ and CD8+ T-cell response to inhibit tumor growth and prevent recurrence in newly diagnosed glioblastoma patients following standard-of-care radiochemotherapy.
