Navtemadlin + cytarabine is an investigational combination therapy being developed by Kartos Therapeutics for the treatment of acute myeloid leukemia (AML). The combination consists of navtemadlin (KRT-232), a potent and selective oral small molecule inhibitor of MDM2 (murine double minute 2), and cytarabine, a standard-of-care pyrimidine nucleoside analog chemotherapy. Navtemadlin functions by disrupting the MDM2-p53 interaction, thereby stabilizing the p53 tumor suppressor protein and inducing cell cycle arrest and apoptosis in p53-wildtype cancer cells. Cytarabine acts by inhibiting DNA polymerase and incorporating into DNA, leading to the termination of DNA strand elongation. This combination is primarily being evaluated in patients with relapsed or refractory AML and those with AML secondary to myeloproliferative neoplasms.
