This is an investigational combination immunotherapy developed by Memorial Sloan Kettering Cancer Center for the treatment of high-risk neuroblastoma. The regimen consists of allogeneic natural killer (NK) cells administered alongside naxitamab (Hu3F8), a humanized monoclonal antibody that targets the GD2 ganglioside frequently overexpressed on neuroblastoma cells. The therapy is designed to enhance antibody-dependent cell-mediated cytotoxicity (ADCC) by providing a potent source of effector cells (allogeneic NK cells) to interact with the antibody-coated tumor cells. The treatment protocol also includes lymphodepleting chemotherapy with cyclophosphamide and the administration of recombinant interleukin-2 (rIL-2) to promote the expansion and activation of the infused NK cells. This synergistic approach aims to overcome the immune evasion mechanisms of neuroblastoma by combining targeted antibody therapy with donor-derived cellular immunity.
