NB004 is an investigational small molecule drug that acts as a pan-PIM kinase inhibitor. It is being developed for the treatment of advanced and metastatic solid tumors in patients who have exhausted standard treatment options. The drug targets the PIM family of serine/threonine kinases (PIM1, PIM2, and PIM3), which are implicated in cancer cell survival and proliferation. Preclinical studies have shown antitumor activity for NB004, including efficacy in non-small cell lung cancer models resistant to KRAS inhibitors when used in combination with sotorasib. The mechanism of action involves inhibition of all three PIM kinases (pan-PIM inhibition), potentially disrupting oncogenic signaling pathways relevant to tumor growth and resistance mechanisms[2][6]. Clinical development is currently at Phase 1.
