Drug intelligence / Profile preview
Nb106 (also known as Syn-Nb-AF106) is a novel anti-fibrin single-chain antibody (nanobody) developed by the Synapse Research Institute and Maastricht University. It selectively targets a species-conserved thrombin-binding site on fibrin (specifically the N-terminal region of the Fβ chains of fibrin monomers, exposed by fibrinopeptide B cleavage) without binding to fibrinogen or thrombin. By displacing thrombin from growing fibrin fibers, Nb106 prevents the lateral expansion of fibrin fibers and renders the displaced thrombin susceptible to inactivation by antithrombin-III. This results in the formation of thin, amorphous, non-fibrillar clots and impairs clot contraction, offering a potential therapeutic approach as an alternative anticoagulant for thrombotic diseases with pathological fibrin formation.
