Overview

NB127 (also known as Naltos Nasal Powder or VRP324) is an intranasal nanoparticle formulation of the small molecule naltrexone, developed for the treatment of Opioid Use Disorder (OUD). The drug utilizes Nanomerics' proprietary Molecular Envelope Technology (MET), which consists of biocompatible polymer nanoparticles designed to encapsulate and deliver drugs directly to the central nervous system via the nose-to-brain pathway. By bypassing the blood-brain barrier and avoiding first-pass hepatic metabolism, NB127 aims to achieve rapid and efficient delivery of naltrexone to the brain. As a mu-opioid receptor antagonist, naltrexone blocks the reinforcing effects of exogenous opioids, thereby reducing cravings and preventing relapse in patients with OUD. The program was developed by Nanomerics and has been out-licensed to Virpax Pharmaceuticals for clinical development.
