Overview

Nb17-nanoCAR is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat multiple myeloma by targeting the B-cell maturation antigen (BCMA). Unlike conventional CAR-T therapies that utilize single-chain variable fragments (scFvs) for antigen recognition, Nb17-nanoCAR employs a single-domain antibody (nanobody or VHH) derived from the Nb17 clone. This nanobody-based approach offers potential advantages including smaller size, higher stability, and potentially reduced immunogenicity compared to scFv-based constructs. In preclinical studies, Nb17-nanoCAR-T cells demonstrated potent anti-tumor activity, specific cytotoxicity, and cytokine secretion (including IL-2, TNF-α, and IFN-γ) comparable to established BCMA-targeting CAR-T therapies like CT103a. The construct is typically generated via lentiviral transduction of primary T cells.
