Drug intelligence / Profile preview
Nb2-MDTCS is an experimental recombinant fusion protein designed to investigate the role of ADAMTS13 in glycoprotein VI (GPVI)-dependent platelet activation. It consists of the inhibitory anti-GPVI single-domain antibody (nanobody) Nb2 fused to the active MDTCS (Metalloprotease, Disintegrin, Thrombospondin-1, Cysteine-rich, and Spacer) domains of the multidomain metalloprotease ADAMTS13. While monomeric Nb2 typically suppresses GPVI-dependent platelet activation, the Nb2-MDTCS construct binds to von Willebrand factor (VWF) multimers via its MDTCS domains, leading to the clustering and activation of GPVI receptors on platelets. This fusion protein retains the proteolytic activity of ADAMTS13 toward VWF and serves as a valuable research tool to explore GPVI-targeted modulation of platelet activation and receptor cross-talk in arterial thrombosis.
