Drug intelligence / Profile preview
Nb62-r-mAb is a brain-penetrant recombinant monoclonal antibody fusion protein designed to inhibit the complement system in the central nervous system. It consists of the complement C7-blocking monoclonal antibody clone 73D1 (grafted onto a modified mouse IgG2a framework with a D265A mutation to ablate Fc receptor binding) fused at the carboxy terminus of one heavy chain to a nanobody, Nb62, which targets the transferrin receptor (TfR). The Nb62 nanobody acts as a molecular shuttle, facilitating receptor-mediated transcytosis across the blood-brain barrier (BBB). By binding to C7, Nb62-r-mAb prevents the assembly of the membrane attack complex (MAC), thereby reducing neuroinflammation, microgliosis, and synaptic loss. It is being investigated for the treatment of neurodegenerative disorders, specifically Alzheimer's disease.
