NBI-1070770 is a novel, selective, orally active small molecule under development for the treatment of major depressive disorder. It functions as a negative allosteric modulator (NAM) specifically targeting the NR2B subunit-containing N-methyl-D-aspartate (NMDA) receptor. This mechanism aims to modulate glutamatergic neurotransmission implicated in depression. The drug was originally discovered by Takeda and is currently being developed by Neurocrine Biosciences, which acquired rights from Takeda. As of early 2024, it is in Phase II clinical trials for major depressive disorder[1][2][3][4][5][6][7].
