NBI27914 is a potent, selective, and non-peptidic small molecule antagonist of the corticotropin-releasing factor receptor 1 (CRF1R). Developed by Neurocrine Biosciences, it exhibits high affinity for the human CRF1 receptor (Ki ≈ 2 nM) and is primarily utilized as a pharmacological research tool to investigate the neurobiological mechanisms of stress, anxiety, and addiction. In preclinical studies, NBI27914 has been shown to attenuate binge-like ethanol consumption in mice by blocking CRF1R-mediated signaling in the central nucleus of the amygdala (CeA). It works by inhibiting the actions of CRF, a key mediator of the body's endocrine and behavioral responses to stress, thereby modulating GABAergic transmission in brain regions critical to the development of alcohol dependence.
