NBL-020 is a fully human monoclonal antibody that targets tumor necrosis factor receptor superfamily member 1B (TNFR2; also known as TNFRSF1B), a member of the TNF receptor superfamily. Developed using NovaRock Biotherapeutics' proprietary AFIS platform, NBL-020 acts as an antagonist to TNFR2, blocking its signaling and thereby modulating the immune response within the tumor microenvironment. By inhibiting TNFR2, which is highly expressed on regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs), NBL-020 reduces immunosuppression and enhances antitumor immunity. Preclinical studies have shown that it can inhibit tumor growth and prolong survival both as a single agent and in combination with anti-PD1 antibodies, including in models resistant to PD-1/PD-L1 inhibitors. The drug is currently being evaluated in Phase 1 clinical trials for advanced malignant solid tumors.
