NBL-025 is a bispecific antibody (BsAb) developed by NovaRock Biotherapeutics that targets both Programmed Death-Ligand 1 (PD-L1) and CD137 (also known as 4-1BB). The molecule is designed to block the PD-1/PD-L1 immunosuppressive checkpoint while simultaneously providing a costimulatory signal to T cells and natural killer (NK) cells via CD137. This dual mechanism aims to concentrate immune activation within the tumor microenvironment, potentially enhancing anti-tumor efficacy and improving the safety profile compared to systemic CD137 agonists. NBL-025 is currently in the IND-enabling stage for the treatment of various solid tumors and hematologic malignancies.
