NBL-028 is a tetravalent bispecific antibody developed by NovaRock Biotherapeutics for the treatment of advanced solid tumors. It targets both human Claudin 6 (CLDN6), a tight junction protein highly expressed in various malignancies but minimally present in normal tissues, and CD137 (also known as 4-1BB), an inducible co-stimulatory receptor on T cells. By binding to CLDN6 on tumor cells and activating CD137 on immune cells, NBL-028 is designed to selectively stimulate the CD137 pathway within the tumor microenvironment, enhancing T cell proliferation, survival, cytolytic activity, and memory formation. This conditional activation aims to redirect immune-mediated killing specifically toward CLDN6-expressing tumor cells while minimizing off-tumor effects. The drug has demonstrated superior efficacy and safety in preclinical studies and is currently being evaluated in phase 1 clinical trials for patients with advanced solid tumors[1][2][4][7].
