NBM-BMX is a selective small molecule inhibitor of histone deacetylase 8 (HDAC8) currently under development by NatureWise Biotech & Medicals. It is primarily being investigated for the treatment of glioblastoma multiforme (GBM), particularly in cases that have developed resistance to temozolomide (TMZ). The drug's mechanism involves the downregulation of the β-catenin/c-Myc/SOX2 signaling pathway and the upregulation of WT-p53 mediated MGMT inhibition, which collectively sensitizes resistant GBM cells to chemotherapy, inhibits cell proliferation, and triggers apoptosis. Clinical studies are evaluating its safety and efficacy as both a monotherapy and in combination with standard-of-care treatments like TMZ.
