NBQ72S is an orally bioavailable, small molecule tyrosine kinase inhibitor (TKI) that selectively targets the Human Epidermal Growth Factor Receptor 2 (HER2). Developed by Nantong Bencao Quadriga Medical Technology, the compound is specifically engineered for high blood-brain barrier permeability to address a significant unmet need in the treatment of HER2-positive breast cancer patients with central nervous system (CNS) involvement. It is primarily being investigated for the treatment of leptomeningeal metastases (LM) and brain parenchymal metastases, where it aims to inhibit the signaling pathways driving tumor growth and survival in the cerebrospinal fluid and brain tissue. By selectively binding to the HER2 kinase domain, NBQ72S blocks downstream oncogenic signaling while minimizing off-target effects associated with EGFR inhibition.
