NBTXR3 is a first-in-class radioenhancer composed of functionalized hafnium oxide nanoparticles. It is designed to be administered as a one-time intratumoral injection prior to standard ionizing radiotherapy. Upon activation by radiotherapy, the unique physical properties of the nanoparticles significantly increase the energy deposited within tumor cells—up to nine times more than radiotherapy alone—leading to enhanced tumor cell death without increasing damage to surrounding healthy tissues. This physics-based mechanism not only amplifies local tumor destruction but also primes an adaptive immune response that may help attack metastatic tumors. The drug has been evaluated in multiple clinical trials across various solid tumors including head and neck squamous cell carcinoma (HNSCC), soft tissue sarcoma, non-small cell lung cancer (NSCLC), pancreatic cancer, rectal cancer, and others[2][5][7][8]. Nanobiotix is the originator and primary developer; Janssen Pharmaceutica (a Johnson & Johnson company) has entered into a global co-development and commercialization agreement for this product[8].
