Overview

NC-6201 is an antibody-drug conjugated micelle (ADCM) developed by NanoCarrier Co., Ltd. for the treatment of EGFR-expressing cancers. It utilizes a micellar nanoparticle platform composed of polyethylene glycol-poly(amino acid derivative) polymers with a diameter of 20-100 nm. The targeting component is NCAB001, an anti-EGFR monoclonal antibody attached to the micelle surface, while the payload is E7974, a novel hemiasterlin analogue that acts as a microtubule inhibitor. This delivery system is designed to enhance the therapeutic index of the cytotoxic payload by improving tumor-specific accumulation and reducing systemic toxicity. Preclinical evaluations have shown significant antitumor activity in EGFR-positive models, including pancreatic and triple-negative breast cancer xenografts, with a favorable toxicity profile in animal studies.
