NC318 is a first-in-class humanized IgG1κ monoclonal antibody developed for the treatment of advanced solid tumors, including non-small cell lung cancer and head and neck cancer. It targets Siglec-15 (S15), a protein expressed in the tumor microenvironment that mediates immune suppression. By blocking S15, NC318 relieves immune suppression within tumors and may enhance anti-tumor immune responses, particularly in patients who do not respond to existing checkpoint inhibitors like anti-PD-1/PD-L1 therapies[3][7][4]. The drug has been investigated both as monotherapy and in combination with pembrolizumab (an anti-PD-1 antibody) to potentially overcome resistance to current immunotherapies[5][2].
