Overview

NC410 is a recombinant fusion protein composed of leukocyte-associated immunoglobulin-like receptor 2 (LAIR-2) fused to the Fc region of human IgG1. It acts as an immune modulator by binding to collagen, thereby blocking the interaction between collagen and LAIR-1 (CD305), an inhibitory receptor expressed on immune cells. This blockade relieves the suppression mediated by LAIR-1, leading to enhanced activation and stimulation of dendritic cells, myeloid cells, and T lymphocytes within the tumor microenvironment. Preclinical studies have shown that NC410 reduces tumor growth in T cell-dependent models and binds specifically to collagen-rich areas where LAIR-1+ immune cells are localized. The drug is being developed primarily for use in combination with pembrolizumab (Keytruda) for advanced or metastatic solid tumors, including colorectal cancer (especially microsatellite stable or low MSI), ovarian cancer, endometrial cancer, gastric cancer, esophageal cancer, head and neck cancers, cervical cancer, lung cancer, and other solid tumors[4][6][3][9].
