Overview

NC525 is a humanized monoclonal antibody targeting leukocyte associated immunoglobulin-like receptor 1 (LAIR-1). It is designed to selectively bind LAIR-1, which is expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs), leading to their direct killing via ligation-mediated apoptosis. Preclinical studies have shown that NC525 inhibits the growth of AML cells, eradicates LSCs, and spares healthy hematopoietic stem cells and other immune cell subsets. The drug has demonstrated efficacy in various AML models, including patient-derived xenografts, and shows potential synergy with standard-of-care therapies such as venetoclax and azacitidine. Currently under development by NextCure for relapsed/refractory AML, myelodysplastic syndrome (MDS), and chronic myelomonocytic leukemia (CMML), NC525 is administered intravenously and is in Phase 1 clinical trials[1][2][3][4][5][7].
