Overview

NCB-0846 is an orally available small-molecule inhibitor of TRAF2- and NCK-interacting kinase (TNIK), a key component of the Wnt/β-catenin signaling pathway. Developed by Carna Biosciences, it binds to TNIK with high affinity (IC50 of 21 nM) and disrupts the formation of the TCF4/β-catenin transcriptional complex, thereby suppressing the expression of Wnt target genes. Preclinical studies have demonstrated its efficacy in inhibiting the growth of colorectal cancer stem cells and preventing metastasis in lung cancer models. Beyond oncology, NCB-0846 has shown potential in treating liver fibrosis and epilepsy, and acting as a radiosensitizer in lung squamous cell carcinoma.
