Drug intelligence / Profile preview
NCC-1566 is a first-in-class, orally active, small molecule inwardly rectifying potassium channel (Kir) activator, specifically targeting Kir4.1 (KCNJ10) channels. Developed by Nissan Chemical Corporation, NCC-1566 is designed to restore potassium and glutamate homeostasis in astrocytes, thereby hyperpolarizing the resting membrane potential and reducing neuronal hyperexcitability. In preclinical rodent models, NCC-1566 has demonstrated potent anticonvulsant and antiepileptic activity, showing efficacy in models of acute seizure, mesial temporal lobe epilepsy (MTLE), amygdala kindling, and sudden unexpected death in epilepsy (SUDEP). Furthermore, NCC-1566 has shown synergistic potential when combined with levetiracetam in pharmacoresistant seizure models, suggesting its potential as an add-on therapy for refractory epilepsy.
