NCD-38 is a small molecule inhibitor specifically targeting lysine-specific demethylase 1A (KDM1A), also known as LSD1. It is primarily investigated for its potential in treating glioblastoma multiforme (GBM) by targeting glioma stem cells (GSCs). NCD-38 works by inhibiting the epigenetic activity of KDM1A, which is often overexpressed in GBM and plays a crucial role in maintaining the stemness of GSCs. Treatment with NCD-38 has been shown to reduce cell viability and neurosphere formation, induce apoptosis, and promote differentiation of GSCs. Mechanistically, it triggers the unfolded protein response (UPR) pathway. In preclinical orthotopic models, NCD-38 has demonstrated the ability to reduce tumor progression and improve survival.
