NCE-052 is a novel small molecule positive allosteric modulator (PAM) of the Kv3.1 (KCNC1) voltage-gated potassium channel. It is being developed for the treatment of epilepsy and seizure control, specifically targeting the restoration of rapid parvalbumin-positive interneuron (PVI) firing. In preclinical studies using *Scn1a* haploinsufficient mouse models, NCE-052 has demonstrated the ability to delay seizure onset and significantly increase the temperature threshold for febrile seizures. It was designed as an analog of existing Kv3.1 modulators to improve upon the properties of tool compounds like AUT1, aiming for potential human therapeutic use in seizure suppression and epilepsy prevention.
