NCI-006 is a small molecule inhibitor of lactate dehydrogenase A (LDHA) developed by researchers at the National Cancer Institute (NCI). It is designed to modulate the tumor microenvironment (TME) by inhibiting lactate fermentation, which reduces glucose consumption, lactate production, and extracellular acidification. These metabolic changes are intended to alleviate the immunosuppressive conditions of the TME, thereby enhancing the activity and viability of immune cells such as CAR-T cells. In preclinical models of neuroendocrine tumors (NETs), NCI-006 has been studied as a monotherapy and in combination with mitochondrial complex I inhibitors like IACS-010759 to prevent metabolic rewiring toward oxidative phosphorylation and maximize anti-tumor efficacy.
