Overview

ncSTING ADC is an antibody-drug conjugate (ADC) developed by Pfizer that utilizes a noncleavable linker to deliver a potent Stimulator of Interferon Genes (STING) agonist payload directly to the tumor microenvironment (TME). By targeting specific tumor antigens, the ADC aims to activate the innate immune response and drive a type I interferon (IFN) response while minimizing systemic toxicity associated with non-targeted STING agonists. Preclinical studies have demonstrated that ncSTING ADCs elicit robust antitumor activity and tumor regression in various murine models, with efficacy influenced by Fcγ receptor binding. The use of a noncleavable linker ensures that the payload is released primarily within the targeted environment, reducing systemic immune activation compared to the administration of the free payload.
