Overview

NCX 1728 is a novel nitric oxide (NO)-donating phosphodiesterase-5 (PDE5) inhibitor developed by Nicox. It represents the lead compound in a new class of molecules that combine NO-mediated effects with PDE5 inhibition within a single molecule. The drug is designed to lower intraocular pressure (IOP) and provide neuroprotection, with potential applications in glaucoma and retinal diseases. Its mechanism involves enhancing ocular perfusion and improving photoreceptor function, primarily through stimulation of the nitric oxide/soluble guanylyl cyclase signaling pathway. Preclinical studies have shown that NCX 1728 can abolish retinal and vascular dysfunction induced by ischemia/reperfusion injury, suggesting promise for treating retinopathies characterized by dysfunctional ocular perfusion and neovascularization[1][2][3][4][5][8].
