NCX 1880 is a des-nitro derivative of a nitric oxide (NO)-donating phosphodiesterase-5 (PDE5) inhibitor developed by Nicox. It has been used as a comparator in preclinical studies evaluating NO-donating compounds for ocular vascular and retinal protection. Unlike its parent compound, NCX 1728, which donates NO and acts as a PDE5 inhibitor, NCX 1880 lacks the NO-donating property due to the removal of the nitro group. In animal models of endothelin-1-induced ischemia/reperfusion injury in the optic nerve head and retina, NCX 1880 did not enhance ocular perfusion or improve photoreceptor function, highlighting the importance of NO donation for therapeutic efficacy in these settings[10].
