Overview

NCX 4251 is a novel, patented ophthalmic suspension of fluticasone propionate nanocrystals developed for targeted topical treatment of the eyelids. Fluticasone propionate is a synthetic corticosteroid with high affinity for the glucocorticoid receptor—approximately ten times greater than dexamethasone—and has been widely used in non-ophthalmic indications such as asthma and allergic rhinitis. NCX 4251 is designed to deliver fluticasone directly to the eyelid margins via an applicator, minimizing intraocular steroid exposure and reducing risks like increased intraocular pressure or cataract formation associated with other topical steroids. The drug is being developed primarily for acute exacerbations of blepharitis and has also shown potential in treating dry eye disease, particularly in subgroups of patients with more severe symptoms. Clinical trials have demonstrated that NCX 4251 is safe and well-tolerated, though efficacy endpoints were not met in all populations; post hoc analyses suggest benefit in certain patient subgroups[2][5][6][7][8].
