Overview

Nd1-Phosphonohistidine (DrugBank ID: DB01899) is an experimental small molecule and a phosphorylated derivative of the amino acid L-histidine. It is characterized by a phosphate group covalently bonded to the N-delta (pi) nitrogen of the imidazole ring. In biological systems, phosphohistidine isomers function as high-energy intermediates in various phosphotransfer reactions, most notably within bacterial two-component signaling systems and in the catalytic cycles of enzymes such as nucleoside diphosphate kinase and succinyl-CoA synthetase. Due to the acid-lability of the phosphoramidate (P-N) bond, this compound is chemically distinct from the more stable O-phosphorylated amino acids like phosphoserine. Currently, Nd1-phosphonohistidine is classified as an experimental compound with no approved therapeutic indications, no active clinical trials, and no identified commercial developers or brand names.
