Overview

The ND5 peptide vaccine is an experimental cancer vaccine targeting the NADH dehydrogenase subunit 5 (ND5) protein, which is encoded by mitochondrial DNA. It was investigated as a potential immunotherapy for renal cell carcinoma (RCC) based on the observation that somatic mutations in mitochondrial DNA can create tumor-associated mitochondrial antigens (TAMAs). In preclinical studies conducted at the University of Pennsylvania, researchers identified ND5 mutations in the RENCA tumor cell line. However, unlike the COX1-targeted vaccine, the ND5 peptide vaccine did not demonstrate significant therapeutic efficacy in protecting against RENCA tumor challenge in experimental models, suggesting that not all TAMAs are equally effective as vaccine targets.
