Overview

ND886 is a novel, selective small-molecule inhibitor of platelet glycoprotein VI (GPVI) currently under investigation for the treatment of acute ischemic stroke (AIS). GPVI is a key collagen receptor on platelets that triggers activation and thromboinflammation. By inhibiting GPVI, ND886 aims to prevent microvascular failure and futile reperfusion—a condition where large vessel patency is restored but microcirculatory flow remains blocked by platelet-neutrophil aggregates and capillary plugging. Preclinical studies indicate that ND886 reduces neutrophil adhesion and preserves microvascular flow without significantly increasing bleeding risk, a common complication with traditional antiplatelet agents. It is being evaluated in a Phase II clinical trial (NCT05836753) for patients receiving reperfusion therapy to improve clinical outcomes by mitigating reperfusion injury.
