NDC-1308 is a novel small molecule drug developed by Endece LLC for the treatment of neurodegenerative and demyelinating diseases. Its primary mechanism of action is as an agonist of estrogen receptor beta (ERβ), which leads to the induction of remyelination and inhibition of demyelination in the central nervous system. This dual action differentiates it from other multiple sclerosis therapies. Preclinical studies have shown that NDC-1308 can directly induce differentiation of oligodendrocyte progenitor cells (OPCs) to promote myelin repair, upregulate genes such as lipoprotein lipase (LPL) and apolipoprotein C2 (ApoC2) in macrophages to shift them toward an anti-inflammatory M2 phenotype, and provide neuroprotection. The compound does not exhibit significant estrogenic activity at therapeutic doses. It has been investigated for multiple indications including multiple sclerosis, Alzheimer’s disease, acute respiratory distress syndrome (ARDS), Niemann-Pick disease type C, neuromyelitis optica spectrum disorder, osmotic demyelination syndrome, COVID-19, and type 2 diabetes mellitus[1][3][4][5][6].
