Overview

NDI-101150 is a novel, oral, highly selective small molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1), a member of the MAP4K kinase family. HPK1 acts as a negative regulator of T cells, B cells, and dendritic cells. By inhibiting HPK1, NDI-101150 reactivates anti-tumor immune responses even under immunosuppressive conditions. The drug has demonstrated dose-dependent pharmacodynamic effects and target engagement in early clinical studies. It is being developed by Nimbus Therapeutics for the treatment of advanced solid tumors—including renal cell carcinoma (RCC), non-small cell lung cancer (NSCLC), and gastric/gastroesophageal cancers—both as monotherapy and in combination with pembrolizumab. Early-phase clinical trials have shown promising anti-tumor activity and good tolerability in heavily pretreated patients with advanced solid tumors.
