NDI-219216 is a highly potent and selective non-covalent small molecule inhibitor of Werner syndrome helicase (WRN), developed by Nimbus Therapeutics for the treatment of advanced solid tumors, particularly those with microsatellite instability-high (MSI-H) status. Preclinical studies have shown that NDI-219216 triggers a DNA damage response in MSI-H tumor cells, suppressing cell viability and promoting cancer cell death. The compound has demonstrated significant tumor regression and complete responses at low oral doses in preclinical models, including those refractory to immunotherapy and chemotherapy. Its non-covalent mechanism may offer more durable on-target activity and a potentially wider therapeutic window compared to covalent inhibitors. As of early 2025, it is being evaluated in a Phase 1/2 clinical trial for patients with advanced solid tumors[1][2][3][4][5].
