Overview

NDI-225673 is an oral, highly selective small molecule inhibitor of salt-inducible kinase 2 (SIK2) being developed by Nimbus Therapeutics (via its subsidiary Nimbus Salacia) for the treatment of inflammatory bowel disease (IBD) and other chronic inflammatory and autoimmune conditions. SIK2 is a key signaling node in myeloid cells that regulates proinflammatory gene expression. By selectively inhibiting SIK2, NDI-225673 suppresses the production of multiple proinflammatory cytokines (such as TNF-alpha, IL-23, and IL-6) while simultaneously upregulating pro-resolution and tissue repair factors (such as IL-10), thereby promoting mucosal healing. NDI-225673 is currently in Phase 1 clinical development.
