Overview

NDL2 is a first-in-class **PD-L1 protein degrader** designed as a **novel bicyclic peptide** therapy for cancer immunotherapy[1][5][6]. Unlike traditional monoclonal antibody checkpoint inhibitors, which block PD-L1 surface interactions with PD-1, NDL2 binds and drives degradation of all forms of PD-L1—including post-translationally modified and resistant variants found in the cytoplasm and nucleus—using the cell’s natural protein disposal machinery[1][5][6]. This strategy targets resistant cancer cell populations that current antibody therapies cannot reach, aiming to overcome immunotherapy resistance and restore durable immune activation in tumors[1][5][6]. Preclinical data indicate efficacy in aggressive triple-negative breast cancer models and non-small cell lung cancer, as both monotherapy and in combination with anti-PD-1 agents[1][6]. No toxicity has been observed in preclinical models so far[1].
