NDM-101

NDM-101 (also known as NT-0167) is a small molecule inhibitor of the NLRP3 (NOD-like receptor protein 3) inflammasome, developed by NodThera. The NLRP3 inflammasome is a multi-protein complex that plays a critical role in the innate immune system by triggering the maturation and release of pro-inflammatory cytokines, specifically interleukin-1β (IL-1β) and interleukin-18 (IL-18). Overactivation of this pathway is implicated in a wide range of chronic inflammatory and neurodegenerative conditions, including Alzheimer's disease, Parkinson's disease, and non-alcoholic steatohepatitis (NASH). NDM-101 was identified as an early lead compound in NodThera's pipeline and progressed into Phase 1 clinical testing to evaluate safety and pharmacokinetics. While it served as a proof-of-concept for the company's NLRP3 platform, NodThera has since advanced next-generation compounds, such as NT-0796 and NT-0249, which offer optimized properties like enhanced brain penetration for central nervous system indications.
