Overview

NDT9513727 is a potent, selective, and orally bioavailable small molecule inverse agonist and competitive antagonist of the human C5a receptor (C5aR1, also known as CD88). Developed by Neurogen, it targets the complement system by binding to an allosteric, extra-helical site on the C5aR1 G-protein-coupled receptor, specifically between transmembrane helices 3, 4, and 5. This mechanism blocks the pro-inflammatory signaling induced by C5a. The compound exhibits high species selectivity for primate (human and macaque) and gerbil receptors, with significantly lower affinity for rodent C5aR1. It has been utilized in research for various inflammatory and neurodegenerative conditions, including sepsis, rheumatoid arthritis, Crohn's disease, and Alzheimer's disease.
